Als Jake Blues uit de gevangenis wordt vrijgelaten, gaat hij met zijn broer Elwood naar het weeshuis waar ze samen opgroeiden. Moeder Overste stuurt hen er op uit om binnen elf dagen 5000 dollar bij elkaar te halen om het weeshuis te redden.
